Using oral swabs, this method involved lysis followed by direct PCR, a nested invasive reaction, and visualization with gold nanoparticle probes, all contained within a closed tube. The strategy behind the genotyping assay is determined by the invasive reaction's proficiency in recognizing a single base. This assay's capacity for rapid and straightforward sample preparation allowed for the detection of 25 copies/L of CYP2C19*2 and 100 copies/L of CYP2C19*3 within a period of 90 minutes. Twenty oral swab samples were correctly typed for CYP2C19*2 and CYP2C19*3, in agreement with pyrosequencing, suggesting the remarkable potential of this method for single nucleotide polymorphism analysis in sample-scarce regions, enabling the application of personalized medicine.
